Abstract (EN)
Glucagon-like peptide-1 (GLP-1) is a peptide with 30 or 31 amino acids. GLP-1 has many metabolic effects such as increasing insulin secretion and slowing gastric emptying. Dipeptidyl peptidase-4 (DPP-4) is one of the peptides that play a critical role in glucose metabolism. DPP-4 enables the breakdown of incretins and increases blood glucose. DPP-4 is involved in the degradation of GLP-1. The nicotine contained in cigarettes is thought to activate the GLP-1 hormone. The aim of the present study was to investigate the changes in blood GLP-1 and DPP-4 levels in rats exposed to cigarette smoke. The data obtained will give an idea about the effect of exposure to cigarette smoke on GLP-1, DPP-4 and insulin levels. In this study, 21 female Wistar albino rats, 2-3 months old and weighing 200-250 g, were used. Rats; control, acute cigarette smoke, and chronic cigarette smoke. Cigarette smoke was administered by inhalation by creating a smoke chamber. Animals in the acute smoking group were exposed to smoke by burning a total of 10 cigarettes. In the group exposed to chronic cigarette smoke, 1 cigarette was burned for each animal twice a day and this process continued for 14 days. After 24 hours, blood samples were taken from the animals. GLP-1 and DPP-4 levels were measured in sera from rats by Elisa method. As a result of the evaluations, it was found that exposure to cigarette smoke increased the average of GLP-1 levels. When acute exposure to cigarette smoke is compared with chronic exposure; It was found that acute exposure increased the GLP-1 level more. There was no significant effect of acute and chronic exposure to cigarette smoke on DPP-4 level. As a result, while serum GLP-1 level was higher in rats exposed to acute cigarette smoke, no significant effect was observed on DPP-4 levels. Keywords: Cigarette smoke, DPP-4, Elisa, GLP-1, Wistar albino rat
