Effect of boric acid on IL-1α, IL-6, TNF-α and Caspase-3 protein expressions in cyclosporine A-induced testicular damage-induced rats

Abstract (EN)
There has been a significant increase in male infertility in recent years. There are many environmental or hereditary factors that negatively affect male reproductive health. One of these is the drugs used in the health sector. Today, thanks to the developing technology and the health and medical industry, organ transplant surgeries are increasing and the success rate is also increasing. However, some problems occur in the reproductive systems of individuals who have organ transplants following their treatments. This situation is due to the use of immunosuppressants such as CsA (Cyclosporin A) for the successful transplantation. CsA creates a toxic effect in the body and causes damage to the reproductive organs and sperm. BA (Boric acid) has a healing effect on many cellular damages due to its antioxidant, anti-inflammatory and anticarcinogenic effects. Our thesis study was conducted to examine the healing effects of BA in rats whose testicular tissue was damaged by giving CsA. Within the scope of the study, MDA (Malondialdehyde), GSH (Glutathione) and CAT (Catalase) analyses were performed and their measurements were carried out with a spectrophotometer. The amount of IL-1α, IL-6, Caspase-3 and TNF-α proteins found in testicular tissue was analyzed with Western blot technique and histopathological measurements were made. As a result of the statistical evaluation of all data; a decrease in the amount of MDA and an increase in the level of GSH and CAT were observed in the testicles of rats given BA. In addition, an increase in caspase-3 protein expression and a decrease in the expression of IL-1α, IL-6 and TNF-α were observed. In the rats given CsA, which is the damage group, a decrease in the levels of CAT, GSH and Caspase-3 was observed, while an increase in the levels of MDA, IL-α, IL-6 and TNF-α was observed. In the testicles of rats given BA + CsA, which is the treatment group, an increase in the amount of antioxidant GSH, CAT and caspase-3 and a decrease in the levels of damage indicators such as MDA, IL-α, IL-6 and TNF-α were observed compared to the damage group. As a result, we believe that BA, which has antioxidant properties, can treat testicular damage and toxicity caused by CsA and prevent possible damage. Therefore, we hope that BA can be studied on humans in the future to provide positive effects on the human reproductive system.
