Protective effect of theranekron against cellular damage of cisplathin on the testis

Infertility is an important health problem that is increasing in men today due to environmental and chemical factors, obesity and chemotherapy. In our study, we aimed to correct the damage caused by cisplatin in the early period in spermatogenic cells in the testis with Theranekron. A total of 30 rats were used in our experiment and the groups were divided into 5 groups, 6 animals in each group, as TH group (0.3mg/kg), CP group (1.5mg/kg), TH+CP group (0.3mg/kg;1.5mg/kg), Sham group, and Control group. Hematoxylin and eosin staining was used to evaluate the morphology of the cells, TNFα, NFkB, IL-1β, IL-6 for inflammation and cell regeneration, and γH2AX immunoreactivity was examined to show early stage DNA breaks. The TUNEL technique was also used to show apoptotic cells. In the CP group, a decrease in markers of inflammation and cell regeneration was observed. High γH2AX immunoreactivity was observed, and an increase in apoptatic cells was also observed. The exact opposite of these statements was observed in the TH+CP group. The TH group showed immunoreactivity close to the Sham and Control groups. These statements were found to be statistically significant (p<0.05). In the CP group, the damage in spermatogenic series cells and the inflammation in the interstitial area were observed to be visibly reduced in the CP+TH group. In the TH group, no morphological change or inflammation was observed in the cells. As a result, it was observed that Therenekron gave positive results in cell damage resulting from early cisplatin application. It was observed that Theranekron played a role in the regeneration of spermatogenic series cells by suppressing inflammatory areas in particular. We think that our study is a good reference for experiments to be conducted especially on the testicles. We think that with the new studies, the usability of Theranekron as a drug especially in male infertility will be demonstrated.
