Investigation of success level of genetic manipulations on antibiotic and enzyme production of streptomyces strains

Abstract (EN)

Soil samples were collected from five different areas of University of Çukurova and 14 different Streptomyces strains were isolated. S35, S47, S48 and S50 of these strains were found to be have an antibacterial activity against test organisms which are Staphylococcus aureus, Bacillus subtillis and Pseudomonas aeruginosa. The antibacterial activity of these three strains were tested at three different medium and these results indicated that different mediums can increase or decrease the ability of antibiotic production of Streptomyces strains. In other research indicated that, the antibiotic production activity is found to be related with having plasmids. The plasmids that isolated from S47 and S48 strains, were transferred to S40 strain which has not antibacterial activity, by electroporation method. 37 of 250 strains that selected from regeneration medium, became having antibiotic production activity, but only 9 of these strains protected this property. In other research, the xylanase activity (at the end of the seventh day) of S40, S47 and S48 strains were found as 50,92U/mL, 49,51U/mL and 47,79U/mL respectively. After doing the protoplast fusion among these strains which have the highest xylanase activity, 9 strains which have different colony morphologies, were selected. The xylanase activity of (40-48) R2 and (47-48) R3 strains were increased in comparison with their parents, by 25,9% and 31,9% respectively. This study also indicated that the enzyme activity can be improved by protoplast fusion technique
