Abstract (EN)
COX, is the key enzyme that catalyzes in the biosynthesis of the prostaglandins from the substrate AA. The two COX isoforms have been identified, COX-1 and COX-2. DM is a group of metabolic diseases characterized by from defects of insulin secretion. The two most common forms of DM are type 1 and type 2 diabetes. RSV which is important antioxidant present that has a variety of biological and pharmacological effectsIn this study, we aim on the basis of knowledge to understand the changes of COX-1 and COX-2 gene expression, protein level and complications of DM in kidney tissue after RSV treatment in STZ induced-diabetic rats44 wistar albino male rats were divided into 6 groups. After 3 days of induced experimental diabetes, blood glucose levels and body masses of the rats was monitored. After the induction of chronic diabetes RSV extract was administered. At the end of 2nd month, all rats were sacrified under anesthesia and their kidneys were removedRenal tissue samples fixed in formaline were embedded in parafine for immunohistochemical study and results evaluated by qualitative method. Quantitative Real time PCR results were assesed by statistical methods. This is the first gene expression study to clarify the relationship between RSV and the expression of COX-1 and COX-2 enzyme in kidney tissue in STZ-induced diabetic ratsIn this study, there were no differences in COX-1 mRNA level among groups. We found to increased gene expression in diabetes group and found statistically differences between control group and diabetes group in terms of COX-2 gene expression (p=0.043). In addition, no significant differences were also found in COX-2 mRNA level of other groups. The findings of the immunohistochemical study indicated that COX-1 expression was inhibited by RSV treatment in sham control+RSV group. However, there were increased COX-1 expression in diabetes+RSV due to the inefficient inhibition. On the other hand, the results of study showed that COX-2 expression was not inhibited by RSV treatment.
