Investigation on the changes of oxidative stress parameters and the effect of safranal in the tissues of rats exposed subchronic thinner inhalation

Abstract (EN)
Thinner, which is used industrially intensively as well as enjoyable and drug abuse, can cause excessive production of free radicals in the body, leading to oxidative stress and cellular damage. The organism balances and removes the harmful effects of free radicals with the activation of enzymatic and non-enzymatic antioxidants result from thinner toxicity. Safranal is one of the components known to have high radical scavenging activity in saffron flowers. In this study, effects on oxidative stress parameters in brain, liver, lung, kidney and testicular tissues of rats treated safranal by using gastric gavage were investigated in rats exposed to subchronic thinner inhalation In this study, rats (Winstar Albino) were divided into 4 groups as follow: Control (K), Safranal (S),Thinner (T), and Thinner+safranal (T+S). Each group was composed of 10 rats and study continued 8 weeks. Thinner inhalation was applied to T and T + S groups in a container with NaOH tablets twice a day. Safranal were given to S and T + S groups by using gastric gavage at a dose of 0.1 mL /kg/day. After 8 weeks liver, lung, brain, kidney and testicular tissues were taken for analysis from experimental animals and the tissues were homogenized and MDA, GSH, NOX, TAS ve TOS levels were detected in the supernatants. The obtained data were analyzed by ANCOVA test using SPSS packet program, p <0.05 significant value was accepted. The lung, liver, kidney, testis and brain tissues which are thinner-inhaled groups were observed oxidative stress table as expected. In the T + S groups, the effect of safranal eliminated or reduced oxidative stres with decreasing of the level MDA and TOS and increasing of the level GSH and TAS. In addition it was showed that the decreasing level of NOx was found statisticaly significant (p <0.05) As a result; It has been found that the protective effect of safranal is not same level in all tissues in the buffering of adverse effects and the prevention or alleviation of possible cell damage Thus, these datas acoording to preventive medicine may contribute on worker's health preventation and the development of new modulations in combat with volatile substances.
