Abstract (EN)
In this study, the growing conditions of sesame plants that have been planted in Kozan, Sarıçam, Karataş, Seyhan districts of Adana, and Tarsus district of Mersin how the Sesame Phyllody disease seen in sesame is affected by the growing conditions, the detection of the disease by PCR in the identified isolates and research on molecular similarity rates were investigated. In each district, 2 or more fields where sesame seeds were cultivated were determined and the fields were checked once a week after the plants were 20 cm tall. During the controls, plants showing symptoms of the disease were collected and brought to the laboratory for analysis. As a result of the observations that have been made, it was observed that Sesame phyllody disease caused less infection in sesame plants which had a better maintenance conditions and vegetatively growed well. Sesame phyllody disease was molecularly determined in the PCR studies of the samples obtained as a result of field controls. Nested PCR method was used to detect Sesame phyllody disease in PCR studies and for this purpose P1/P7 and R16F2n / R16R2 primers were used in this study. The presence of Sesame phyllody disease in 10 out of 40 samples collected were determined by PCR method as a result of laboratory studies. A total of 3 samples, including one sample from Kozan-Sarıçam, one sample from Karataş-Seyhan and one sample from Tarsus, were sent to the sequence analysis and the isolates obtained in this study showed 96% similarity with other domestic and foreign isolates.
