Molecular investigation of in vitro cytotoxic effects of crownvetch (Coronilla varia) and bird's-foot trefoil (Lotus corniculatus) plant extracts on breast cancer cells

Abstract (EN)

Cancer development is caused by a series of events involving genetic events, angiogenesis, metastasis and mutations. In this study, plasmid DNA protection activities, apoptotic, autophagic and telomerase gene activities and anti-metastatic activities of aerial parts of two plant species Lotus corniculatus and Coronilla varia EA, MeOH and water extracts on MCF-7 and MDA-MB-231 cells were investigated. Apoptosis was determined in MDA-MB-231 cells treated with Lotus corniculatus EA extract. In addition, it was found that in the MCF-7 cells, treated with Lotus corniculatus EA extract, the expression of Bcl-2 gene was increased and the expression of autophagy genes was increased. Results were associated to phenolic contents in extracts determined by Dionex Ultimate 3000RS UHPLC analysis. In Lotus corniculatus EA, linoleamide substance was detected and it was confirmed that anticancer results were related with this substance in literature review. In addition, after treatment of MCF-7 cells with Coronilla varia EA and MDA-MB-231 cells with Coronilla varia water, Bax gene expression increased and Bcl-2 gene expession decreased due to apoptotic mechanism. In this study, it is predicted that linoleamide will contribute to the comprehensive studies by making drug resistance reversible for being the potential raw material of the drug.
