Title: Expression of heat shock protein HSP 70 in recurrent pregnancy loss. Student Name: Taner ÖZEL Supervisior: Assoc. Prof. Dr. Tamer ZEREN Depertment: Physıology Depertment Aim: Recurrent pregnancy loss is a health issue with a high prevalence. Heat shock proteins play a crucial role in the formation of the quaternary structure of proteins, intracellular balance of free radicals, and cellular defense against stress factors. Our aim is to investigate the expression levels of HSP 70 protein, one of the heat shock proteins, in the abortus material of patients experiencing recurrent pregnancy loss and to normalize it with the reference protein beta-actin. Material and Method: 24 volunteer patients were divided into three groups: "healthy non-pregnant individuals (Group 1)", "healthy pregnant individuals (Group 2)," and "individuals with recurrent pregnancy loss (Group 3)." Abortus materials from Groups 2 and 3 and endometrial biopsy samples from Group 1 were obtained. Western Blot analysis was conducted on the collected samples, and the expression levels of HSP 70 and beta-actin proteins were individually examined, followed by the normalization process. Results: There has been a decrease in the expression level of the HSP 70 protein in Group 3 patients (p<0.05). An increase in the expression level of the beta-actin protein has occurred in Group 3 patients (p<0.05). When the normalization process (HSP 70 / Beta-Actin) was conducted, a decrease (p<0.05) was observed in Group 3 patients. Conclusion: A decrease in HSP 70 protein levels and an increase in beta-actin protein levels have been observed in recurrent pregnancy losses. Statistical analyses have revealed significant results in patients belonging to the recurrent pregnancy loss group. The lower expression of the HSP 70 protein in comparison to the control group in other groups suggests that it could be one of the etiological reasons for pregnancy loss.
