Protective and thereupatic effects in testis tissue of apocynin on testicular ischemia-reperfusion injury in a rat

Abstract (EN)

Purpose: This experimental study was designed to explore the protective effect of apocynin, NADPH-oxidase inhibitor, on testis damage induced by I/R in rat model. Methods: Thirty-two rats were randomly divided into a control group and three I/R groups (4-h ischemia followed by 1-h reperfusion). The first group was named control, control-operation. In the second group, there become torsion and detorsion. İn the third group in addition to the procedure applied saline (10 ml/kg i.p. ) in the second group after was conducted at the 210. minutes of ischemia. In the fourth group in addition to the procedure applied apocynin (20 mg/kg i.p. ) in the second group after was conducted at the 210. minutes of ischemia. The histopathological findings, including tubule diameters, giant cell, degenerated cell, poured into the lumen of seminiferous tubules epithelium, interstitial hemorrhage and also tissue malondialdehyde (MDA), superoxide dismutase (SOD), catalase (CAT), glutathion peroxidase (GPX), reduced glutathione (GSH), myeloperoxidase (MPO), oxidative stress index (OSİ), total oxidative capasity (TOC), total antioxidan capasity (TAC) levels were determined. Results: Malonoldialdehyde, TOC and OSI levels are significantly higher in I/R and I/R+SF group than Control group. Superoksiddismutaz, CAT, GPX and GSH levels are significantly lower in I/R and I/R+SF group than Control group. Malonoldialdehiyde, TOC and OSI levels are significantly lower in I/R + Apocynin group than I/R and I/R+SF group. SOD and CAT levels find significantly higher in I/R + Apocynin group than I/R and I/R+SF group. Myeloperoksidase level was find lower in I/R + Apocynin group than I/R and I/R + SF groups. TAC level was find higher in I/R + Apocynin group than I/R and I/R + SF groups, but didn?t find significantly difference for istatistcially. In the groups that were applied I/R it was found that the tubule diameters got smaller but other values significantly sinificantly got higher. However in the group given apocynin treatment the average diameter of seminifer tubules significantly rise were found when compared to the I/R, I/R+SF groups. Conclusion: The beneficial effects of apocynin on testiculer I/R injury were evaluated for the first time. Key words: Testis, ischemia-reperfusion injury, apocynin
