DNA interaction and antimicrobial activity study of thiosemicarbazone derivatives

The chemotherapy for cancer disease has had enormous developments and researchers still as yet trying to develop new and more effective drugs to fight this illness. Thiosemicarbazone and Thiosemicarbazides take possession of a wide range of biological applications. The interaction of three different isatin thiosemicarbazone zinc complexe with calf thymus DNA were examined by using UV spectrophotometry studies. Interaction of 1H-Indole-2,3-Dione-3-(N-Phenyl Thiosemicarbazone, 1H-Indole-2,3-Dione-3-[N-(4 Nitrophenyl)- Thiosemicarbazone and 1H-Indole-2,3-Dione-3-(N-Benzyl Thiosemicarbazone) zinc complexes with CT-DNA was carried out by increasing concentration of DNA from 0 to 30μl by using UV spectrophotometer study. These studies confirmed that, the complex binds to DNA through the minor groove according to the docking studies, in addition, all molecules were used to carry out DNA binding study with DNA (downloaded from Protein Data Bank) using Discovery Studio 3.5 to obtain the most favored binding mode of the ligands within the DNA. Isatin thiosemicarbazones derivatives and its metal complexes were evaluated in vitro against 16 types of bacteria and one type of fungi. Results shown that the studied compounds have different antibacterial activities against five types of tested bacteria, and it may have a good synergism with ceftriaxone against some types of bacteria which tested in vitro. According to the results obtained it was observed that metal complexes might had a heightened activity compared to the ligand against some types of tested bacteria.
