Determination of some viral factors potato production areas in Tokat province

Abstract (EN)
In the scope of present study, potato leaves and tuber samples, showing virus like symtoms, were collected from potato production areas in Centrum, Niksar, Erbaa, Başçiftlik and Artova districts of Tokat. Total 418 leaves and 91 tuber samples were tested with two step polymerase chain reaction (PCR) using virus-specific primers to Potato virus Y (PVY), Potato virus S (PVS), Potato virus X (PVX), Potato leafroll virus (PLRV). Single or dual infections of these viruses have been determined in 220 (%52.63) leaves samples and 68 (%74.72) tubers samples. 197 (%47.12) out of 418 leaves samples were infected with PVY, 70 out of 418 samples with PVS (%16.74), 25 of samples with PVX (%5.98) and 22 of samples with PLRV (%5.26). 67 (%73.62) out of 91 tubers samples tested resulted with positive with PVY, 6 out of 91 samples with PVS (%6.59) and 1 of samples with PLRV (%1.09). Sequence analysis of some pozitif isolates was obtained and compared with isolates which recorded in Genbank for phylogenetic analysis tree.
