Abstract (EN)

TMZ is an anti-ischemic and anti-anginal agent. Scientific studies have shown that trimetazidine is mitochondrial stabilizer used for indications such as dizziness, tinnitus and also shown that in cases of hypoxia or ischemia prevents decreasing of intracellular level of ATP by blocking the oxidation of fatty acids. Generally, substances are known protecting the cells against hypoxic conditions, may show some neuroprotective effect. In sensorineural disorders such as tinnitus, beneficial effects of TMZ was noticed, unlikeon the central nervous system, such as movement disorders, which is on the side effects of neuroleptic drugs with similar side effects were observed.In this study, potential protective and toxic effects of TMZ on neurons and glial cells, the morphological changes of cells evaluated by MTT assay to compare rates of vitality, for the determination of rates of apoptosis and apoptotic markers by fluorescent staining was investigated by comparing the amounts of caspase-3. To form oxidative stress and thus degeneration in cells, we used H2O2 and MSG that causes excitotoxicity with excessive excitation.As a result, we found that toxic dose of trimetzidine is very high, and trimetazidine significantly protects neuronal and glial cells against cell death induced by H2O2 and MSG. Because of these findings, we suggest that trimetazidine may participate in the treatment options for neurodegenerative diseases.Key words: Trimetazidine, Neuron, Glial, Cell Culture, Apoptosis, MTT
