Abstract (EN)

The desired goal of surgical periodontal therapy is to provide periodontal regeneration. Many materials and bioactive agents have been developed for this purpose. Platelet-Rich Fibrin (PRF) is a bioactive material containing platelets in high concentrations obtained from the patient's own blood, which has been of interest to clinical investigators in recent years. A number of studies have been done on the production of PRF but no standardization of g force and centrifugation time has been achieved. The aim of this thesis is; to compare the amount of growth factors and cytokine obtained by equalizing the g values of an ordinary laboratory centrifuge with the L-PRF device, to determine the optimal g force value and the centrifugation time by the amount of the growth factors. Periodontally and systemically healthy 10 subjects were included in the study. From every patient; A total of 13 tubes of blood were collected, including 1 tube control group (L-PRF, 400×g 12 min) and 12 tube study groups (200, 400, 600, 800×g; 10, 12, 14 min). Blood tubes were centrifuged at specified time and g forces and PDGF-AB, TGF-β1, IL-1β release on days 1, 3, 7 were examined. For the three molecules, the highest release values on 1st day were seen in the control group. On the 3th and 7th days, the release values from the control group were decreased, whereas the higher release values were seen in the groups of PDGF-AB and IL-1β at 800×g for 14 min and 600×g for 10 min and for TGF-β1 at 400×g 12 min. These results demonstrate that standard laboratory centrifuges may be an appropriate alternative to L-PRF device and may provide growth factor/cytokine release for longer periods of time.
