Investigation of the effect of boric acid combined with platelet rich fibrin on the difference and vitality of dental pulp stem cells

Abstract (EN)
The aim of this study was to evaluate the biocompatibility and compare the potential in vitro cytotoxic effects of boric acid and injectable platelet-rich fibrin on human dental pulp stem cells (DPSCs) and their effects on odontogenic differentiation of DPSCs for dental tissue engineering. Human dental pulp tissue were isolated from extracted impacted third molars from healthy adult patients aged from 18 to 25 years. Injectable platelet-rich fibrin (I-PRF), 1 µM concentration of boric acid and a combination of them were prepared and applied to the cells by direct contact method. The viability of the stimulated cells with experimental material and DMEM control group were evaluated on the flow cytometry device with Anneksin-V and Propidium iodide at the end of the 24th, 48th and 72nd hours. To evaluate the odontogenic differentiation capacity of both cells, Alizarin-red (ALR) staining were performed. Two-way analysis of variance was used to compare flow cytometry values, and multiple comparisons were made with the Tukey HSD Test. When the data were evaluated, I-PRF group had the highest mean live cell value in terms of viable cell viability. In terms of cell viability findings, there is a statistically significant differences were noted between the sampled hours (p> 0.05). There was no significant difference between the groups in terms of early apoptosis findings (p>0.05). There is a statistically significant difference between the groups in terms of late apoptosis findings and the lowest late apoptotic cell value belongs to the I-PRF group. (p<0.05). In terms of necrotic cell findings, boric acid had the lowest necrotic cell value and no significant difference was observed between the groups (p>0.05).The best odontogenic differentiation potential on DPSCs was determined in the group combined with boric acid and I-PRF. In this study, I-PRF and boric acid were demonstrated for biocompatibility and odontogenic differentiation, and additional animal and clinical studies are required to further explore their potential for use in dental tissue engineering.
