Abstract (EN)

Platelet rich plasma (PRP) is used at widely varied branches such as cardiovascular surgery, orthopedics, urology, maxillofascial surgery, dentistry, sports medicine, plastic surgery right now. Due to having such a broad spectrum of usage, researches for efficiency, standardization and preparation of PRP are still going on for the moment. In this study, our aim was to prepare PRPs from two different donors and apply it to fibroblast cell cultures at the same concentrations to see its effects. For this purpose we took whole blood samples from two different donors and prepared the PRPs with a two staged centrifuge. By dividing each sample to four different groups with different concentrated PRPs, we were able apply PRPs to eight different cell cultures (J1, J2, J3, J-PPP, E1, E2, E3, E-PPP). Furthermore for both donors cell cultures without PRPs were established (J1ext, J2ext, J3ext, E1ext, E2ext, E3ext). We applied the time as 24 hours for our study. While interpreting the results, we have considered the increase in mitotic activity, measurement of the levels of "Fibroblast Growth Factor (FGF)", "Insulin Like Growth Factor (IGF)", "Vasculer Endothelial Growth Factor (VEGF)", "Transforming Growth Factor (TGF)", "Platelet Derived Growth Factor (PDGF-AB)" and whether these levels were corelated with PRP concentrations or not. At the end of the study compared to the control group, we have determined statistically significant increase in mitotic activity, in other words proliferation at each and every group expect j-ppp and e-ppp. Due to fact that both j-ppp and e-ppp were poor in terms of platelet concentration, increase in proliferation was not meaningful when compared to the control group. When growth factors were examined, PDGF-AB was the factor corelated with the increase of mitotic activity. Increase in other factors were not corelated with proliferation. PRP; is an autologous blood product which is easily obtainable, with a low side effect profile. At the field of plastic surgery it is used especially for its positive effects at wound healing. It contains the key elements of wound healing such as platelets and growth factors at a high concentration. In our study we have concluded that PRP causes a statiscally significant increase in proliferation at fibroblast cell cultures. We have seen corelation between increased levels of PGDF-AB with proliferation. By preparing equally concentrated PRPs from two different donors we have seen similar effects on proliferation. This made us think that while evaluating the efficieny; concentration is more definitive than platelet count. On the other hand, in order to make our study a reference point for those who are preparing their own PRPs, and in order to understand efficieny of PRP further detailing and research must be done.
