Association of Aurora-a gene polymorphisms with lung cancer in Turkish population

Abstract (EN)

Lung cancer is the leading cause of cancer mortality in the world, accounting for %16 of all cancers and %28 of all cancer deaths (35% men, 19% women). There are several environmental and genetic factors which could be changed in each individual, effect lung cancer risk. The genomic alterations can exist in molecular pathways, such as regulation of cell cycle. The Aurora-A kinase which is a member of protein kinase family, regulates the functions of centrosomes, spindles and kinetochores required for proper mitotic progression. The kinase function can change with the F31I and V57I polymorphisms in Aurora-A gene. Our aim is to reveal the polymorphic variations of codon 31 and 57 in Turkish population and find out the relation with lung cancer. Included in this study, 86 incident lung cancer cases who are newly diagnosed and 102 age frequency-matched, non smoking, no familial cancer history, randomly selected community controls were studied to determine Aurora-A polymorphisms. The genomic DNA was extracted from whole blood. After the CR reaction all samples were genotyped with DNA sequence technique. The frequencies of both genotypes (he31 lie, Val57lle) in lung cancer patients were not significantly different from those in controls. earson's x2 p values were found 0,430 and 0,73, respectively (p>0.05). The present study is the first Turkish data which reveals the relation between Aurora-A polymorphisms and lung cancer risk. The results of our and many other studies contribute to the efforts to understand the association between lung cancer and Aurora-A polymorphisms as well as to the prospective diagnosis and treatment phases.
