Investigation of proteome profiles and androgen receptor, PSA, RNASEL gene polymorphisms related to prostate cancer in Turkish population

The early detection of cancer is crucial for its ultimate control and prevention. Proteome and genome studies provide data and new insights into cancer diagnosis, following the prognosis and treatment plans. Proteomic analysis of cancer provides new opportunities to identification of candidate biomarkers.The aim of this study is to determine protein profilles of prostate cancer patients in Turkish population, identify multiple biomarkers in serum and investigate related frequent gen polymorphisms in prostate cancer.Proteom profilles were analyzed for 47 prostate cancer, 50 benign prostate hiperplazi and 20 post prostatectomy PSA value 0 patients and 20 healthy control group by surface enhanced laser desorption/ionization time-of-flight (SELDI-TOF) mass spectrometry. Molecular genetic analysis were carried out towards the polymorphisms of Androgen Receptor (AR) CAG repeats, Prostate Specific Antigen (PSA) promoter region and RNASEL gene.Seven serum differential proteins were identified in the PCa group compared with the control groups (P < 0.01) in proteome analysis. There were 4 proteins at the lower serum level and 3 proteins at the higher serum level in the PCa group. 6628 Da was evaluated as special and privileged for prostate cancer (p<0.001). According to the molecular genetic analysis there were no statistical difference between prostate cancer and benign prostate hyperplasia groups due to the polymorphisms of (CAG)n repeats in AR gene ekson 1 region. PSA -158, PSA -252 and RNASEL D541E.It is possible to identify different biomarkers for prostate cancer by means of proteomic studies. In this sectional study, a new serum biomarker (6628 Da) of PCa for Turkish population is remarkable. The results show that there is no significant relationship between AR gene short repeats and polymorphisms of promoter region of PSA gene and RNASEL D541E. Further studies should be performed to clarify the proteome and genotype association. Results obtained from this study show that proteomic researches will provide a highly accurate and innovative approach for the early diagnosis of cancer.
