Relation between radiographic evaluation for hip and elbow dysplasia with FCI and IEWG criteria and DNA based predictive blood tests in some dog breeds in Turkey

Association of hip and elbow dysplasia in some dog breeds in Turkey with radiographic evaluations and DNA-based predictive blood test results according to FCI (Fédération Cynologique Internationale) and IEWG (International Elbow Working Group) criteria In this study, it was aimed to investigate the gene polymorphisms that may be effective in the development of hip and elbow dysplasia, which is one of the developmental orthopedic diseases in dogs, and to evaluate the potential of using the genetic information obtained at an early age in detecting dogs with the disease in susceptibility to the disease. In this context, radiographical findings of diseased and healthy animals were compared with the genotypic dataset of four selected polymorphisms. Thirty (n=30) German Shepherd and 30 (n=30) Labrador Retriever dogs were included in the study. 36 of these dogs were female and 24 were male, all of whom had reached bone maturity at the age of 1 year. Pelvis and elbow radiographs were taken in appropriate positions under anesthesia after lameness and orthopedic examination performed on dogs presented for routine screening to get breeding permission. Radiographs were evaluated and graded according to FCI and IEWG criteria. Following DNA isolation from blood samples, genotyping was performed using Real Time PCR technology. The obtained data were analyzed statistically. According to statistical evaluations, there was a statistically significant difference between males and females in dogs with only hip dysplasia. (p=0.0036) A statistically significant difference was found between SNP1 numbered (rs8897564) close to the SMYD3 gene location in dogs evaluated as having both hip and elbow dysplasia (p=0.015; p=0.007, respectively). In conclusion, polymorphisms occurring close to gene locations determined to be effective in hip/elbow dysplasia were evaluated for the first time in this study, and single nucleotide mutation (SNP1) with the number (rs8897564) occuring close to SMYD3 gene location was found to be significant for both hip and elbow dysplasia. This polymorphism, which was first evaluated in elbow dysplasia, will shed light on future studies.
