Project Title: The Comparison Of Reproductions Of Isolates, Obtained In Turkey, With Cutaneous Leishmaniasis In Liquid Culture Media Student Name: Yalçın DEMİR Advisor: Prof. Ahmet ÖZBİLGİN Master of Science: Medical Parasitology M.S Objective: Ascertain the liquid culture medium reproducing Leishmania promastigotes, to be used in molecular studies, isoenzyme analyses and drug studies, at most and within the shortest time. Materials and Methods: Obtained from the patients with cutaneous leishmaniasis in Turkey, Leishmania isolates stored in liquid nitrogen were vitalized in an appropriate occasion and inoculated into NNN culture medium. The isolates reproduced in the culture medium were genotyped via the method of Real-Time PCR with primers and probes specific to ITS1 sequence. The isolates, obtained from the patients with cutaneous leishmaniasis in Turkey and determined as L. tropica, L. major and L. infantum. /donovani after genotyping, were inoculated into RPMI-1640, M199, Schneiders, Nutrient Broth and Brain Heart (infusion) Broth culture media. Inoculated culture media were controlled on every day up to 12th day after inoculation and reproduction density of those was assessed in Thoma slide. Results and conclusion: It is observed that L. tropica, L. major and L. infantum/donovani isolates, obtained from the patients with cutaneous leishmaniasis in Turkey, can be reproduced in RPMI-1640 culture medium better than M199, Schneiders, Nutrient Broth, Brain Heart Broth culture media, used in the study, with much more promastigote reproduction for all Leishmania spp. beginning from 8th day (p<0.05). Therefore, it was ascertained that the best liquid culture medium is RPMI-1640 if promastigote reproduction of Leishmania spp., causing CL, obtained from Turkey is needed at most. Later on, it was concluded that M199, Schneiders, Brain Heart Broth and Nutrient Broth mediums would be suitable. Key Words: Leishmania, Culture media, Turkey, Culture.
