Determination of genetic diversity of bluefish (Pomatomus saltatrix) in Turkish coasts

Abstract (EN)
While detailed molecular studies have not been found on Bluefish (Pomatomus saltatrix) populations, which have high commercial value in our country, there are few studies in the world. In the proposed thesis, it is aimed to determine the genetic structure of the bluefish, which is an economically valuable species distributed in the seas of our country, by using microsatellite markers and marker genes using DNA sequencing method. The study (Hopa, Trabzon, Giresun, Samsun, Sinop, Kefken, Istanbul, Igneada, Erdek, Çanakkale, Bodrum, Izmir, Mersin and Adana) will be carried out on samples obtained from a total of 14 stations. DNA extraction was performed at Trabzon Fisheries Central Research Institute Molecular Genetics Laboratory, amplified by PCR technique using fluorescent marked primers and microsatellite loci (8 loci), mtDNA genes (COI and 16S regions) and nuclear gene (Rhodopsin) universal primers, and sequenced in ABI 3500 device and sequenced. microsatellite fragment sizes will be determined separately. Using the obtained data, DnaSP 4.0 program will create intra-species, inter-population and intra-population haplotypes. Haplotype diversity (Hd) value, Nucleotide diversity (Pi, ) and nucleotide diversity (k) values will be calculated, phylogenetic relationships between species and a phylogenetic tree will be created with Mega5, allele sizes and frequencies for each species and locus will be determined with Genmaker, Arlequin and Calculation of molecular variance components and gene flow between populations will be made with Amova, and basic criteria for population genetics will be tested by using Genalex 6 to determine the relationship between genetic difference and geographic distance. From these data, population genetic parameters, phylogenetic and phylogeographic structure of Bluefish (Pomatomus saltatrix) will be revealed by using appropriate genetic markers (microsatellite and sequence sequencing), primarily by comparing closely related species and their populations with Genbank data.
