Molecular characterization of the laurel (Laurus nobilis L.) genotypes collected from different regions of turkey

Abstract (EN)

This study was carried out to make molecular characterization of 94 Laurel genotypes from 26 provinces where the Laurel plant is widely grown in Turkey. In this study, 16 ISSR and 10 SCoT primers were used in the research laboratory of Field Crops Department of Abant İzzet Baysal University in Bolu. Of 373 bands obtained in ISSR analysis, 348 were found to be polymorphic and the polymorphism rate was 94.04%. According to ISSR results, Nei genetic distances ranged between 0.17 and 0.70 and the mean value was found to be 0.39, The most distant genotypes were 56th (Zonguldak/Ereğli/Gülüç) and 10th (Antalya/ Gazipaşa/ Demirtaş) genotypes with a value of 0.70. Genotypes with the closest genetic distance with value of 0.10 were identified as 56th (Zonguldak/ Ereğli/ Gülüç) and 42nd (Sinop/ Türkeli/ Center) genotypes. In the SCoT analysis, 175 of the total 227 bands were found to be polymorphic and the polymorphism rate was determined as 76.07%. According to SCoT results, Nei genetic distances ranged between 0.12 and 0.51 and the mean value was found to be 0.26. The most distant genotypes were 44th (Kastamonu/ Abana/ Center) and 14th (Antalya/Kumluca/Center) genotypes with a value of 0.51. The genotypes with the closest genetic distance with a value of 0.12 were identified as 62nd (Istanbul/ Sile/ Center) and 50th (Bartın/ Amasra/ Kaleşah) genotypes. In UPGMA cluster analysis of 94 laurel genotypes, ISSR and SCoT results were compatible with each other and genotypes were divided into 2 main groups. As a result of ISSR and SCoT analysis, a similar polymorphism distribution was observed and the correlation coefficient (r) was found 0,88 in the correlation analysis performed on Nei genetic distances to determine the linear relationship size of both analysis results. It is thought that the findings will compose resources for advanced breeding techniques and contribute to the preservation of genetic diversity and management of genetic resources for the country's economy.
