Genetic analyses of brown meagre (Sciaena umbra Linnaeus,1758) and shi drum (Umbrina cirrosa Linnaeus,1758) distributed along Turkish coastal waters

To analyze filogenetic and population structure of shi drum and brown meagre, 254 fishwere sampled from eight locations of Turkish coastal. The sequence analysis of Cyt-b, 16SrRNA and RFLP analysis of Cyt-b, 16S rRNA and ND1 gene sites were examined tofilogenetic and population structure of these fish. Based on sequence analyses, 25 and 20haplotypes were specified for brown meagre and for Shi Drum in Cyt-b gene and 4haplotypes for brown meagre and 8 haplotypes for shi drum in 16S rRNA. Nucleotidediversity in 16S rRNA and Cyt-b genes were found 83.8% and 52.6% for brown meagreand 80.5% and 73.6% for shi drum, respectively. Molecular variances analysis showed thatFST value was found 0.07364, Cyt-b molecular variances analysis for shi drum indicatedthat FST value was 0.1881 (P<0.001). The calculated nucleotide diversity (Pi) anddivergence (k) for brown meagre and shi drum were between 2.97% and 2.87% for Cyt-bgene and between 1.54% and 0.61% for 16S rRNA gene, respectively. Based onphylogenetic analysis no differentiation was achieved at the level of genus. According toRFLP analysis results, no polymorphic structure was found in population level.Key Words: Umbrina cirrosa, Sciaena umbra, Cyt-b, haplotype, RFLP, nucleotidediversities
