Determining of genetic diversity of barley varieties grown in Turkey using ISSR (inter simple sequence repeat) marker technique

Abstract (EN)

This study was conducted to determine genetic diversity in 28 barley cultivarsoriginated from Turkey (22 two-raw and 6 six-raw) using some physio-morphological characters and ISSR DNA molecular markers. To study somephysio-morphological characters, these varieties were planted in field during 2003 -2004 growing year. Based on this study, no any differences found among the barleyvarieties.To study genetic diversity in 28 barley varieties by ISSR molecular marker,64 ISSR primers were screened in 7 varieties. 57 of 64 produced a PCR product. 10ISSR were selected according their polymorphism level. These ISSR primers totallyproduced 75 bands, and 51 were polymorphic. Average ?Polimorphism InformationContent? (PIC) was 0.611, with minimum PIC 0.354 and maximum PIC 0.844.Genetic Distance Coefficient average was found 0.214, and range from 0.05 to 0.37.Minimum genetic distance was found between Efes-2 and Yesevi-93 and maximumwas between Karatay-94 and Aday-4. In this research, ?physio-morphologiccharacteristics? was found insufficient for detecting similarities and differencesbetween barley genotypes where ?ISSR Markers Technique? was successful forseperating the cultivars.Key Words: Barley, Hordeum vulgare, ISSR markers, Genetic Diversity
