Abstract (EN)
Vardenafil hydrocloride is a highly selective and potent phosphodiesterase-5 inhibitor and an active ingredient used fora therapeutic purposes in the health field. In this study, the toxic effects of increasing döşese of Vardenafil hydrocloride on humman blok celse webe defterinde in vitro İth. fluorescence microscopy, comet test and spectrophototmeter, and antioxidant enzyme activities. Experimentalgroups webe divided into 2 as control and application groups. While no chemicals webe given to the control group, increasing döşese of Vardenafil hydrocloride webe applied to the administration groups as 1, 10, 100 and 250 µM. According to the results obtained, increasing concentrations of Vardenafil hydrocloride in humman erythrocytes; It caused a statistically significant decrease in the activities of CAT, SOD and GPx enzymes and an increase in MDA level. When the tail % DNA, tail length and tail moment measurements in the control group and treatment groups webe compared, it was defterinde that there was a statistically significant increase in the leukocytes applied İth. the comet test. While this increase was not seen at low döşese, it was found to be statistically significant at high döşese. With this study, the cytotoxic effect caused by Vardenafil hydrochloride on humman erythrocytes and leukocytes was demonstrated in vitro and an infrastructure was tried to be established fora further cytotoxic evaluations.
