Effect of vildagliptin supplementation on pancreas islet cell

Abstract (EN)

The aim of this study is to investigate the effect of orally administration of vildagliptin on isolated pancreatic islet cells in rats. Pancreatic islet cells were isolated after vildagliptin (100 mg/kg/day) adminstration for 4 days.The isolated Pancreatic islet cells were incubated within 0 and 300 µM H2O2 containing medium at +4°C for 15 min. Then, the islets cells viability were examined with fluorescein diacetate (FDA) and propidium iodide (PI) mixture by using a fluorescence microscope. A number of islets were stored for glucose stimulation test, lipid peroxidation, protein oxidation and Superoxide dismutase (SOD) activity assesment. The dose of Vildagliptin (100 mg/kg/day ) were statistically different when compared to control (P<0,05). Vildagliptin (100 mg/kg/day) administration protect the cells aganist the H2O2 induced lipid peroxidation and proteion oxidation. Glucose release was observed variable in the islet cells isolated from rats by vildagliptin administration.Finally, vildagliptin (100 mg/kg/day) administration ameliorate the H2O2 induced damage in terms of the islet cells viability and oxidative parametersApplication of vildagliptin to the donor before isolation process may be suggested to protect islet cells from early damage. However, further investigation is needed to improve the isolation procedure and cell viability before transplantation.
