Abstract (EN)
Today, the potential risks of animal products make microbial resources produced by biotechnological methods attractive. The disadvantages of traditional sources and methods used in the production of chondroitin sulfate, which is used in various applications in medicine, veterinary, pharmacy and cosmetics sectors, have made microbial production an important alternative. In our study, the effect of the vgb gene on the production level of microbial chondroitin sulfate produced by our team was discussed. In this study, E. coli pETM6-PACF-vgb+ strain was formed by transferring (pETM6-PACF-vgb+) plasmid to non-pathogenic E. coli bacteria, and E.coli pETM6-PACF bacteria with plasmid pETM6-PACF without the vgb gene were used for chondroitin production. In the last stage, sulfation was applied to chondroitin using the selective sulfation method, and chondroitin sulfate was produced. The molecular structure of the produced Chondroitin Sulfate was proven using HPLC and NMR analyzes. Our study proves the supportive effect of the vgb gene on bacterial growth characteristics and also reveals the advantages of strains containing the vgb gene for MCS production.
