DETERMINA TION OF B10CHECIMAL VARI A TION IN WILD SPECIES OF WHEA T (M.Sc Thesis) Semiha GENÇALİ ERİŞEN GAZİ UNIVERSITY INSTITUTE OF SCIENCE AND TECHNOLOGY January 1998 ABSTRACT The objective of this study was to determine within and among population variation for two of each Ae. squarrosa, Ae. speltoides spel., Ae. speltoides lig. and T.dicoccoides populations collected from Ceylanpinar State Farm in Şanlıurfa in South Eastern Anatolia. For that purpose the populations were screened in terms of 3 isozymes Ep-1, Amp-1, Amp-2 and 2 seed storage proteins Gli, Glu covering 8 loci of the wheat genome. For isozymes IEF, for gliadins A-PAGE and for glutenins SDS-PAGE techniques were used. The analyses of the populations collected from Gökçayır Saraççeşme and Gürgürbaba locations were resulted in high level of within and among population variations. For that reason these locations were suggested as Gene Management Zones (GMZs) in the content of the project. As a result of comparison with the cultivated varieties, the variation determined in this study can be used as a genetic resource in breeding studies for improving varieties having high yield, quality and resistance to diseases.
