Comparison of microrna profiles in infants born with and without assisted reproductive techniques

Abstract (EN)

Although assisted reproductive techniques have become a routine treatment in human infertility, the cause of perinatal, natal and postnatal problems in newborns born with ART has not been fully explained. In infants born with ART, cardiac, neurological, musculoskeletal system anomalies related to the urogenital system are higher than infants born without ART. In assisted reproductive technology, oocyte and embryo development varies dramatically according to natural pregnancy. Some epigenetic factors permanently alter the germ cells and the embryo's gene expression regulatory system. An important factor in epigenetics is miRNA. MiRNAs are RNAs that are not small encoded and regulate gene expression. Numerous disease-related miRNA changes have been reported in the literature. The aim of the study is to show that the miRNA profile of infants born with or without ART is different and whether this profile is related to the congenital anomaly. 21 babies born using ART and 18 babies born without ART were included in the study. The physical examination findings of the patients were recorded. Venous blood samples were taken from the patients for the miRNA end. Target miRNAs were identified among infertile-associated miRNAs. Our target miRNAs: miR 17-5p, miR 21-5p, miR 23a-3p, miR 92a-3p, miR 141-3p, miR 145-5p, miR 191-5p, miR 483-3p. As a reference miR 16-5p was used. In infants born using ART, we found that miR-145 increased significantly (p <0.05). When we compared the ART group in themselves, we found that miR-145 and miR-483 increased significantly at birth using ICSI (p <0.05). When compared with infants born using ART in terms of anomaly of the control group, we found that this rate was high and significant in the ART group (p <0,05). We could not find a link between the presence of congenital anomaly and the target miRNA profile. As a result, children born using ART should be closely monitored throughout their lives for the diseases associated with miR-145 and miR-483. It is possible in the future that miRNAs can be used as biomarkers. For this, there is a need for further work by parents to examine miRNA profiles.
