Investigation of the effect of horizontal platelet-rich fibrin on bone healing in rat calvariale defects

Abstract (EN)

Objective: The aim of our study was to evaluate the bone healing and regenerative effects of platelet concentrates and to investigate the effect of Horizontal Platelet Rich Fibrin (H-PRF) and Leukocyte and Platelet Rich Fibrin (L-PRF) materials on experimentally created critical size defects. Materials and Methods: In our study, 24 young-adult female Wistar albino rats, 2-3 months old and weighing 200-400 grams, were used. Group 1 was the control group and no procedure was performed. In Group 2, only a critical size defect was created in the skulls of the rats and no biomaterial was used. After the critical size defect was created in the rats in Group 3, L-TZF was applied. After the critical size defect was created in the rats in Group 4, Y-TZF was applied. All rats in the group were sacrificed by high-dose anesthesia 35 days after the operation and tissue samples taken from the skull were sent for histological examination. The results obtained were evaluated statistically and the study was concluded by making comments on the data. Results: The findings obtained showed that in the immune expression of ONC molecule, G2 group was statistically significantly lower than G1, G3 and G4. In addition, the immune expression of ONC molecule was statistically significantly higher in G3 group compared to G4 (p < 0.05). In the immune expression of BMP and ALP molecule, G2 group was statistically significantly lower than G1, G3 and G4 (p < 0.05). No statistically significant difference was found between G3 and G4 groups. In addition, in Hematoxylin-Eosin staining, regenerative tissue formation was significantly more evident in G3 and G4 microscopically compared to G2, and G3 was superior to G4. Conclusion: When the histopathological and immunohistochemical findings we obtained were evaluated, positive effects on bone healing were observed in the L-TZF and Y-TZF applied groups. Among the osteogenic markers we evaluated in bone healing, only ONC expression was found to be superior to L-TZF Y-TZF, and apart from that, no significant difference was found between L-TZF and Y-TZF. We believe that the number of osteogenic markers and subjects evaluated in bone healing should be increased in order to obtain more detailed results on the subject.
