Highly organised organisms which shows complex structures for growing and developing, need communications between cells. In plants , plant growth regulators provide this communication. Cytokinins are nitrogen purine base derivatives that, accelerates plant growth and development. High performance liquid chromatography is the most common separation type which is used in chromatographic separation methods. In recent years , the stationary phases which is called polar embedded and generally includes groups like amide , cabamate and sulfonamide , gave better results in separation of basic analytes instead of conventional C18 columns and moved one step forward about separation superiority. In this study, cytokinin examples was tried to separate with using SP1 amide ?bonded polar embedded stationary phase which was synthesized in Ph.D.Thesis of Aral (2013) earlier. In the study, with using some parameters like mobile phase , pH, temperature some optimizations studies were made. Also , a quantitative study was prepared with natural plant exract that we had. In the study , some validation parameters like LOD ,LOQ were applied and proved that column can be applied the quantitative determination of the various natural analytes
