Investigation of the effects of new phthalocyanine compounds on Alzheimer's disease

Abstract (EN)

In this study, the aim was to investigate the potential of water-soluble peripheral tetra-({6-[3-(diethylammonium)phenoxy)hexyl}oxy substituted zinc and nickel phthalocyanine (DE-C6-ZnQ and DE-C6-NiQ) compounds as drugs against Alzheimer's disease. In this context, the inhibitions and kinetics of the compounds on acetylcholinesterase and butyrylcholinesterase enzymes were determined using UV-Vis spectrophotometry. The plasmid pBR322 DNA nuclease properties of the compounds were investigated using agarose gel electrophoresis. Molecular docking studies on the inhibition of cholinesterase enzymes by the compounds were also conducted. The cytotoxic activities of the compounds against the human neuroblastoma cell line (SH-SY5Y) were examined using the [3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide] (MTT) cell viability test. According to the molecular docking results, it was found that DE-C6-CN had a stronger affinity with AChE than BuChE. DE-C6-ZnQ and DE-C6-NiQ exhibited higher inhibitory effects compared to the standard galantamine, and both compounds showed competitive inhibition on AChE and BuChE according to Lineweaver-Burk plots. DE-C6-NiQ was found to have a stronger cytotoxic effect on SH-SY5Y cells compared to DE-C6-ZnQ. All these results indicate the potential of DE-C6-ZnQ in the treatment of Alzheimer's disease.
