Abstract (EN)
Naphthalimide derivatives are aromatic heterocycles of pharmacological importance as they provide core scaffolding for antitumor, antiprotozoal, antibacterial and antiviral agents. Thanks to its ability to bind to DNA, it acts by showing an antiproliferative effect. In recent years, many literature summaries have been prepared and the structure-activity relationships of naphthalimide have been examined. In the studies, it was understood that the naphthalimide ring system was connected by intercalating between DNA base pairs and various activities were obtained with the substitutions made from various points of the ring. In this study, 2-(2-amino-4-bromo-pyridine)-1H-benzo[de]isoquinoline-1,3(2H)-dione (G1) and the cationic compound (G2), which we obtained by alkylating over pyridine imine, was synthesized. Characterization of the obtained compounds was performed by FT-IR, 1H-NMR, 13C-NMR, ESI-MS analysis methods. DNA interactions have been studied. DNA interactions of compounds were studied by UV-Vis titration, fluorescence, competitive fluorescence, viscosimetry and agarose gel electrophoresis methods. As understood in the results of the study, the interaction of G1 compound with DNA is weak and they probably act as partial intercalation. However, it has been determined that the cationic compound (G2) intercalates with DNA in studies with spectrophotometric methods. Compounds did not damage plasmid DNA in agarose gel electrophoresis.
