Antiproliferative effects of new synthesized flavonoid derived compounds on jak/stat pathway in lung adeno carcinoma cells

Abstract (EN)
Cancer causes the death of one fifth of people in developed and developing countries of the world. The most common types of cancer are prostate, breast, lung, colon / rectum and cervical cancers. Cancer occurs as a result of gradual accumulation of mutations. These accumulated mutations cause resistance to programmed cell death, induce angiogenesis, maintain proliferative signaling, avoid growth suppressors, and potent metastasis potential. What is desirable and ideal in lung cancer chemotherapy is that drugs used only affect cancerous tissue (malignant cells) and have the least toxic effect on healthy cells. One of the important pathways involved in the pathogenesis of cancer is the JAK / STAT signaling pathway, consisting of Janus kinase (JAK) and signal transducers ve activators of transcription (STAT). Flavonoids are thought to have anticancer antithrombotic, antibacterial, antifungal, antiviral and protective properties against cardiovascular diseases. Changes in the chemical structure of molecules can radically alter the biological effects of molecules. The toxic effects of existing chemotherapeutic agents have created a requirement for the synthesis of less toxic but also more effective compounds. The aim of this thesis is to investigate the antiproliferative effects of some newly synthesized flavonoid derived molecules via STAT3 pathway in lung adeno carcinoma cells (A549) and to investigate the mechanisms of STAT3 pathway controlled by these molecules. In this study, after the A549 cells covered the surface of the culture dishes at a rate of 70-80%, SF (serum free medium; serum free) medium was added to the cultures instead of the medium containing 10% FCS. Cells were incubated for 24 hours under these conditions. The newly synthesized flavonoid derivative molecules were then treated with various concentrations (1-100µM). MTT analysis and gene expression analysis with qRT-PCR were performed. As a result, in our study, it has been shown that 5M compound selected among 4 synthetically produced flavonoid derivative compounds has antiproliferative effect at 50µM, 75µM and 100µM concentrations in A549 cell line. It was concluded that the antiproliferation effect did not take place via the JAK / STAT signal pathway, but it could be possible through other signal pathways as MAPK.
