Determination of antibacterial activity of gold (Au) nanoparticles using plant extract with green synthesis method

Abstract (EN)

In this thesis, the synthesis studies of gold nanoparticles (AuNP) were carried out with green synthesis method by using the extract of dried cherry stalk (Cerasus avium L.) without using any other reducing and/or stabilizing agents. The UV-Vis spectrometer, Transmission Electron Microscope (TEM), X-Ray Diffractometer (XRD) and Fourier Transform Infrared Spectrophotometer (FTIR) methods were used in the characterization studies of AuNPs obtained under the most appropriate synthesis conditions. The optimum synthesis conditions of AuNPs were optimized for the extract concentration, added metal solution concentration and synthesis time parameters, and in this context, the optimum extract concentration, the optimum Au solution concentration and the synthesis time were found to be 1.5%, 10-2 M and 60 minutes, respectively. In the application step of this thesis study, the antibacterial effects of the synthesized AuNPs were investigated for gram positive Enterococcus faecalis ATTC29212, Listeria monocytogenes ASKK472, Bacillus cereus ATTC 11778, Staphylococcus aureus ATTC 43300 and gram negative Salmonella Typhimurium ATTC 14028, Enterobacter aerogenes ATTC 13048, Klebsiella pneumoniae, Escherichia coli 0157:H7, Salmonella Enterocolitica, Escherichia coli 0157:H7 ATTC 43895, Escherichia coli ATTC 35150 bacteria. In addition, the catalytic activities of the obtained AuNPs were also determined by carrying out the color removal studies of methyl orange (MO) and methylene blue (MM) dyes.
