Investigation of effects extract obtained from horse chestnut (Aesculum Hippocastanum L.) added to high protein diet on bone and calcium metabolism in rats

The present thesis was planned to determine to effects of extract obtained from horse chestnut (Aesculum hippocastanum L.) seeds given especially on bone and calcium metabolism in feeded rats with high protein. Material of animal, aged 2-3 months, weighting 173-287 g,50 male Sprague-Dawley rats were used. Rats divided into 5 groups as a control and four study groups in each group will have 10 units in and were placed unable to contact each other in the five compartments. While control group was feed control rat diet, I. Study group was fed with added %25 casein to standart diet, II. Study group was fed with added %25 casein to standart diet and extract of horse chestnut, III. Study group was fed with standart diet and extract of horse chestnut, IV. study group was fed with standart diet and 20 ml %50 ethyl alcohol and water mixture.Horse chestnut extract which dissolved 20 ml %50 ethyl alcohol and water mixture was given 100 mg/day/kg to animals via gavage. Rats were housed so as to12 hours light and 12 hours dark at normal room temperature for 30 days. At the end of the study; calcium, parathyroid hormone (PTH), vitamin D, calcitonin, ostecalcin, phosphorus, alkaline phosphatse (ALP), total protein, urea nitrogen and glocose level were determined in were taken blood samples.Appications of high protein diet and horse chestnut extract had no effects on serum calcium levels in contrast with these parameters were obtained a decrease in statiscal significance (P< 0.001) in control and study groups by application of ethanol. In our study according to the findings there aren?t significant differences (P> 0.10) in the levels of PTH, calcitonin, ostocalcin and ALP. Appications of high protein diet had no effects on vitamin D levels, appications of high protein diet with horse chestnut extract and application of ethanol causing a significantly increase (P< 0.07) in vitamin D levels. In our study serum phosphorus levels were elevated in feeded rats with high protein compared to the control group (P < 0.001). In animals which feeded with ethanol total protein levels showed a statistically significant decrease compared to the feeded animals with high protein. Our results show high protein added group as causing an increase (P< 0.001), horse chestnut added groups as causing a decrease (P< 0.001) in plasma urea nitrogen levels. In our study according to the findings application of ethanol was elevated plasma glucose level significantly (P < 0.001).It was concluded, high protein feeded and ethanol extract of horse chestnut had no effect on blood calcium level and hormones which regulate blood calcium levels in contrast with blood calcium level were obtained a decrease, horse chestnut feeded causing a decrease in blood urea nitrogen and will reduced the negative effect of ethanol.
