Abstract (EN)

In the first part of the study, the electrochemical determination of the anticancer drug 6-thioguanine (6-TG) and the solution-phase interaction of 6-TG with DNA oligonucleotides were investigated using disposable pencil graphite electrodes (PGE) in the solution 6-TG. Before and after the interaction, the electrochemical signals of 6-TG and DNA were measured by PGE and the interactions (decrease/increase) were determined using differential pulse voltammetry (DPV) technique based on the changes observed at the signals. In addition, the interaction of 6-TG with DNA interaction at the PGE surface was investigated using electrochemical impedance spectroscopy (EIS) technique. In the second part of the study, the electrochemical detection of 6-TG and the detection of 6-TG-DNA interaction at the electrode surface were examined using single wall carbon nanotube (SWCNT) modified pencil graphite electrodes (SWCNT-PGE). The microscopic surface characterization of the developed nanomaterial based biosensor was performed by scanning electron microscope (SEM). The electrochemical signals of 6-TG and DNA were measured using PGE and SWCNT-PGE. Differential pulse voltammetry (DPV) and electrochemical impedance spectroscopy (EIS) techniques were used for the determination of 6-TG and its interaction with DNA by SWCNT-PGE. In this study, the electrochemical analysis of 6-TG by using disposable graphite electrodes, the electrochemical determination of 6-TG interaction with DNA was performed by using these fast, selective and sensitive protocols. Thus, the results showed that the analysis of 6-TG and the detection of 6-TG and DNA was performed by using these single-use electrochemical DNA biosensors developed in the sduy.
