Abstract (EN)

PRP is obtained by centrifuging whole blood. Because it contains growth factors intensively, it is used widely in many therapeutic areas in recent years. Since PRP is obtained autologously from individual's own blood, its application also prevents the formation of allergenic effects. Mesenchymal stem cells (MSCs) are stem cells having multipotent differentiation ability. since they are originated from mesoderm layer, they have the potential of transforming into mesoderm-derived cells such as condroblast, osteoblast and adipocyte easily. In our study, after getting ethical committee approval, AD-SCs were purified from lipoaspirate/lipectomy material taken from the voluntary donors who have applied to Ege University Faculty of Medicine, Plastic and Reconstructive Surgery for the liposuction procedure. After cultivation AD-SCs were differentiated into adipocytes. PRP was obtained by centrifuging the peripheral blood samples obtained from the same donors. During cultivation and differentiation of AD-SCs into adipocytes, PRP derived autologously from human donors were used in order to supply the growth factors required instead of using fetal bovine serum (FBS) or fetal calf serum (FCS) of animal origin. Thus, it was aimed to prevent or reduce immün rejection or allergenic reactions that may ocur because of animal-derived material. During the differentiation process of AD-SCs into adipocytes, the use of PRP instead of animal serum shortened the differantiation period approximately two-fold. At the same time, it was observed that the amount of cells in the culture medium with PRP was nearly two times greater than the cells in the medium with animal serum. For example, it was determined that dt (doubling time) was 20.087 hours in 20% PRP used culture medium, while dt was 35.99 hours in the 20% FBS used one. In conclusion, it was determined that using PRP instead of FBS in culturing of MSCs was more effective. Since immun rejection can be prevented or lessened by using autolog human material instead of animal derived material, in requirement of FBS or FCS for culturing or differantiation procedures of MSCs in clinical trials the use of autolog PRP can be conveniently recommended.
