Abstract (EN)

Peripheral nerve damages are seen commonly after accidents, sharp objects injuries and particularly as a result of complete transection, delayed and incomplete surgery; they are dramatic views result in progressive muscle atrophy and ineducate functional recovery.Recently, new methods have started to take place besides the different surgery techniques for peripheral nerve repairing in literature.In this context, Stem cells applies have become one of the most popular topics of medicine recently, and hundreds of studies are being published every year.Stem cells, because of their pluripotentiality and unlimited capacity for self-renewal, project great promise for tissue engineering and are expected to allow significant advances for distinct reconstructive procedures.In our study, the effects of the green fluorescent protein (GFP) marked adipose derived mesenchymal stem cells (ADSCs) application were aimed research by functional analysis, histomorphometric and biochemical evaluation on experimental sciatic nerve injury to peripheral nerve regeneration.Experimental 4 Wistar groups (n:8) was designed: A nerve defect of 1 cm was constructed in the left sciatic nerve of the rats was reconstructed with ADSCs application in epineural cover built out of first group.Functional recovery was evaluated by walking track analysis. Histomorphometric studies of the sciatic nerves and gastrocnemius muscles were also performed in postoperative fourth month.In conclusion; All clinic, biochemical and histomorphometric analysis showed that ADSCs application has a possitive effect on supporting peripheral nerve regeneration, stem cells neural differantiation, axone production and myeline formation. This effect is probably due to the nörotrophic factors that are released from the ADSCs during activation. Further study will help to clarify the relative advantages of ADSCs applications on peripheral nerve regeneration.
