Abstract (EN)
Because inadequte perfusion of body part and tissue due to ischemia arterial or reduction of venous blood flood, it is defined that the body part and tissue mentioned are empty of oxygen. Reperfusion of ischemiac tissue causes a crucially damage in comparison of damage happened with ischemia paradoxically in the tissue. Many mechanism play an important role in the death of cell that caused by both regeneration of cell and a new blood flood which is required to be cleaned up toxic metabolite after ischemia. Ischemia/Reperfusion-induced lung injury is a common situation occurring in lung transplantation. Apoptosis (programmed cell death) also known as suicide of cells is physiological event. It is playing a key role in embriyonic development and maintenence of adult tissues.In our study, levels of the genes of SMAD-1, Bmp-2, Bcl-XL, b-FGF, Caspase-3, TGF, PDGFR have been researched for lung tissues which were after the model ischemia/reperfusion (I/R) that created in lung. For that, 40 wistar albino rats were divided equally. Group I= Control group, Group II= İschemia for 60 minutes + reperfusion for 0 hour, Group III= İschemia for 60 minutes + reperfusion for 2 hours, Group IV= İschemia for 60 minutes + reperfusion for 4 hours determined as groups. RNA isolation, cDNA synthesis and RT-PCR (Reverse transcription polymerase chain reaction) were made. Lung tissues were also histopathologically examined.According to the results of gene levels, Caspase-3 (p<0,0001), Bcl-XL (p=0,0011) and TGF (p<0.0001) are statistically significant decrease in groups (p<=0,05). b-FGF (p<0.0001) gene is statistically significant increase (p<=0,05). Bmp-2 and PDGFR genes aren?t statistically significant showed.
