Determination of genetic diversity in Anatolian Buffalo populations grown in Aydın

Abstract (EN)
This study was carried out to determine the genetic diversity of Anatolian Buffalo populations in Aydın province. This study is the first genetic study in Anatolian water buffalo populations in Aydın province. In the study, saliva and spare hair samples were collected from Anatolian Buffalo populations bred in Aydın province, Bozdoğan Ziyaretli, Efeler Çiftlikköy and Söke Serçin regions, and DNA isolation was carried out in the laboratory. Six of the primers (CSSM033, ILSTS033, CSRM060, CSSM045, CSSM057, CSSM057, ILSTS030) for buffalo were studied by FAO/ISAG and mirosatellite analysis was performed. Analyzes revealed a total of 41 alleles. The number of alleles at loci ranged from 4 (ILSTS033) to 11 (CSSM045). It was observed that the observed and expected heterozygosity values varied between (0,560 and 0,629) and (0,568 and 0,651), respectively. The FIS values of the breeds in the study were found to be between 0,0654 and 0,0983. The FST values were between 0,0123 and 0,0321 in three different buffalo population. According to the Nei's Standard Genetic Method, populations Söke-Efeler the populations with the highest genetic distance value (0,3%) within the race. The lowest genetic distance value (0,12%) among the populations was observed in Bozdoğan-Söke populations. According to the DA Genetic Distance Method, the highest genetic distance value was observed in Söke-Efeler population (0,42%) and the lowest genetic distance value was observed in Söke-Bozdoğan population (0,16%). In the study, it was concluded that microsatellite loci can be used to make genetic comparisons of buffaloes. The results obtained are similar to previous studies. It can be a resource for future work.
