Abstract (EN)
The aim of this study is to investigate production possibilities of some rootstock candidate wild plum genotypes obtained by selection breeding using in vitro shoot tip culture. Four selected clone rootstock candidate genotypes belonging to Prunus spinosa, Prunus domestica, Prunus divaricata and Prunus cerasifera, which are selected from Kahramanmaraş and give positive results in scion propagation experiments, were used as material in this study. At the same time, standart rootstocks Myrobolan 29C and GF 677 were used as the control. Fresh shoots of selected genotypes to obtain sterile plants were cultured in Murashige and Skoog (MS) medium. In regeneration stage, five different MS media including four different growth regulators [Thidiazuron (TDZ), 6-Benzylaminopurine (BAP), Gibberellic acid (GA3), and Indole-3-Butyric Acid (IBA)] at various doses (0, 1, 1.5 mg/l; 0, 1, 1.5 mg/l; 0.2 mg/l, and 0.2 mg/l, respectively) were used with their combinations. In the regeneration step of the study, parameters such as percentage of plant development, shoot number per explant and infection rate were examined in both sub-cultures. Subsequently, in vitro plants obtained from the regenerated explants were transferred to two different rooting media including different combinations of TDZ (0.1 mg/l), BAP (0.1 mg/l), and IBA (1 mg/l) and their rooting rates were determined. In this context, generally; it was observed that the genotype 'KLDİ-47' and medium 3 gave better results in terms of shoot number and plant growth rate. Medium-based reaction of genotypes was observed. It was noted that the rooting rates were quite low in the study. Key Words: Plum, rootstock, TDZ, BAP, shoot tip
