Abstract (EN)

Carbonic anhydrases (E.C. 4.2.1.1.) are family of zinc metalloenzymes which were purified from mamalian erythrocyte for the first time. Carbonic anhydrases catalyze the reversible hydration of carbon dioxide in a two-step reaction to yield bicarbonate and proton. The isoenzymes of the enzyme in human eye are CAII and CA-IV. Carbonic anhydrase inhibitors, which reduce aqueous production with a corresponding decrease in intraocular pressure (IOP), have been used as ocular hypotensive agents for the treatment of glaucoma.Previous studies have shown that fungal exstracts have beneficial effects against eye diseases. They also reduce blood sugar and cholestrol levels. They have antioxidant and antitumor effects. In this study, effects of Lentula Edodes, Lactarius Deliciosus, Lactarius Deterrimus, Terfezia Boudieri Chatin fungi on the inhibition of the enzyme carbonic anhydrase have been investigated.First, carbonic anhydrase enzymes from human erythrocyte were purifed using affinity gel and subsequent experiments were carried out with these enzymes. Later, the obtained mushroom extracts on the inhibition of human erythrocyte HCA I and HCA II enzymes were studied. In these studies, the hydrates and esterase activites were taken into account to determine the carbonic anhydrase activities.After studying inhibition effects of the mushroom extract on hydratase and esterase activities of human erythrocyhte HCA-I and HCA-II, % activity graphs were plotted.Key Words: Glaucoma, Carbonic Anhydrase Inhibitors, Lentinula Edodes, Lactarius deterrimus, Lactarius Deliciosus, Terfezia bouderi chatin, mushroom.
