In vitro micropropagation of some prunus rootstocks and evaluation of resistance for root-knot nematodes

Abstract (EN)

In this study, it was targeted to micropropagate in vitro condition of peach, nectarin, GF-677 and Myrobalan 29 C rootstocks by means of shoot-tıp culture. Besides, In vitro grown Garnem, Cadaman, and Myrobalan 29C were tested for resistance to root-knot nematodes, M. incognita race 2 and M. javanica. MS and modified MS (MST) media containing of different concentrations of BAP (0, 0.5, 1.0, 1.5, 2.0 mg/l) with a 0.05 mg/l concentration of GA3 were used in vitro clonal propagation of rootstocks. MS and MST media suplemented with different concentration and combination of IBA and Fe-EDDHA were used for rooting of shoots. The highest number of adventitious shoots for Garnem, GF-677, and Cadaman were obtained on MST medium with 2.0 mg/l of BAP. The best shoot formation for Myrobalan 29 C was obtained on MS medium supplemented with 1.0 mg/l of BAP. The best root formation for GF-677, Garnem , and Cadaman was found on MST medium containing 1.5 mg/l of IBA and 100, 500, 500 mg/l of Fe-EDDHA respectively. Potted plants were inoculated with 1000 second stage of juveniles (J2) of M. incognita race 2 and M. javanica. Three months after inoculation, the ratio of galling in the roots and the number of nematod in the soil of infested rootstocks were recorded. Reproduction ratio of M. incognita race 2 and M. javanica was calculated as 0 on all tested rootstocks. When gall ration was evaluated, gall index of Cadaman was 1 and the gall index of the rest of rootstocks was 0.Key words: Micropropagation, Meloidogyne spp., PCR, Prunus rootstocks, Resistance
