Molecular characterization of some sour orange (Citrus aurantium L.) genotypes with iPBS (inter primer binding site) markers

Citrus fruits are one of the largest fruit groups cultivated in the world and have a high economic importance. They include economic species such as orange, lemon, grapefruit, mandarin and sour orange. Therefore, investigating the genetic basis of citrus species and determining their phylogenetic relationships constitute the most important step of breeding studies. Molecular marker systems offer significant advantages for determining phylogenetic relationships and genetic similarities between genotypes. iPBS marker systems are one of the reliable marker systems developed in recent years and used intensively in the field of molecular breeding. iPBS marker system is a system based on transposon elements in the genome and allows obtaining detailed information about the genome. Therefore, in our study, it was aimed to investigate the phylogenetic relationships of 34 citrus (22 sour orange, 12 other species) genotypes and determine their genetic origins by using iPBS marker system. For this purpose, 28 iPBS markers were used and 437 precise and reproducible DNA band profiles were produced from all primers. Therefore, the average polymorphism rate was 99.04%. PAST 4.2 package program was used in the analyses. As a result of the analyses, principal component analysis (PCoA) of the data was performed and a dendogram of genotypes was created. When the results were examined, it was seen that iPBS markers contributed to the determination of genetic similarities and differences between citrus genotypes.
