Abstract (EN)

In the scope of the study, in order to obtain haploid plants belonging to C. annuum L. species of 4 different varieties (12-081 (Kapia A), 10-714 (Kapia B), 15-416 (Dolma 39), 12-013 (Dolma 47)) In tissue culture studies, the maltose and sucrose effect and pre-temperature applications as a carbohydrate source in food medium and androgenesis in pepper was investigated. In addition, it is aimed to reveal the effect of genotype and different nutrient media on anther culture. Anthers used in the study were subjected to 2 different pre-treatments (25 and 35 ° C (dark)) and cultured in a combination of 2 different sugars (maltose and sucrose). In the present study, Murashige and Skoog (1962), the main nutrient medium and 0.1 mg / l BAP + 4 mg / l NAA + 2.5 mg / l activated carbon, were used as the nutrient media. Callus formation of cultured anthers in pre-treatment and sugar-containing nutrient media were evaluated. When the results of the study were evaluated on the basis of varieties, callus formation was observed in all varieties. When the results were evaluated in terms of environments and pretreatments, the most successful results in terms of callus formation were obtained from pretreatment of maltose as a carbohydrate source for 6 days at 35 ° C. In nutrient environments where sucrose was used as a carbohydrate source, some successful results were obtained depending on the varieties. When the cultivar x medium interaction was examined, the most successful results in terms of callus formation were obtained in Kapia B and Kapia A cultivars in maltose medium which was kept at 35 ° C for 6 days. As a result, this study showed that the use of maltose as a carbohydrate source in pepper anther culture studies had a positive effect on the formation of callus at 35 ° C for 6 days.
