Investigation of the mutagenic effects of some substances used in plant growth with salmonella ames microsome test

Abstract (EN)

In this study, the mutagenic effects of the four different plant growth regulator (Gibberellic Acid, Kinetin, Chlormequat chloride, 2,4,6-trichlorobenzoic acid) that is commonly used in agricultural sector were investigated. Experiments were done with Salmonella Ames Mikrozom test method in the absence (S9-) and presence (S9+) of metabolic activation enzymes. Salmonella typhimurium TA 98 and TA 100 strains of were used in the test system. TA 98 was used for determination of the mutagens that cause of frameshift mutations and TA 100 was used for the mutagens that cause of base-pair mutations. Before starting to mutagenicity tests, the cytotoxic doses of the test substances were determined. Then, 5 non-cytotoxic dose for each test substances were selected. For the determination of mutagenicity of Gibberellic acid 50 µg/plate, 100 µg/plate, 500 µg/plate, 1000 µg/plate, 5000 µg/plate doses were studied. 2.5 µg/plate, 25 µg/plate, 250 µg/plate, 1000 µg/plate and 2500 µg/plate doses of Kinetin substance, 100 µg/plate, 200 µg/plate, 400 µg/plate, 800 µg/plate, 1600 µg/plate doses of Chlormequat chloride substance were studied. 5 µg/plate, 50 µg/plate, 500 µg/plate, 1000 µg/plate, 2500 µg/plate doses of 2,4,6-trichlorobenzoic acid substance were studied. Colony counts obtained test plates were evaluated by compare with spontaneous control plates. According to findings, 2500 µg/plate dose of Kinetin has been found mutagenic on the strain TA 98 in the presenceand in the absence of S9 enzyme. Again, 2500 mg/plate dose of Kinetin was determined to be mutagenic on TA 100 strain in the presence of S9 enzyme experiments. The other doses of Kinetin substance were not observed back mutation on the TA 98 and TA 100 strains. 2500 µg/plate dose of 2,4,6-trichlorobenzoic acid were determined to be mutagenic on strain TA 98 in the absence of S9 enzyme experiments. The other doses of 2,4,6-trichlorobenzoic acid substance were not observed back mutation on the TA 98 and TA 100 strains. The other two substances all tested doses of Gibberellic acid and Chlormequat chloride identified as non-mutagenic. Therefore, for these two substances should be considered that are not caused by mutations such as frameshift and base pair changes.
