Abstract (EN)

In this study; It was aimed to screen the halophilic bacteria isolated from Canik Saltern for cellulase enzyme. It was also aimed to determine the effect of different media components on enzyme activity. In addition, classical and molecular characterization of the cellulase-positive isolates was performed. Halophilic medium containing 20 % NaCl was used for isolation of halophilic bacteria. For the detection of cellulase enzyme, halophilic agar containing 1% carboxymethyl cellulose (CMC) was used. The isolates were intensively planted on this medium and allowed to incubate at 35 ° C for 48 hours. After incubation, they were washed with 1M NaCl solution after 15 minutes by adding kongo red dye to cover the surface of the plates. The formation of clear zones around the biomass was evaluated as positive for cellulase enzyme. According to colony morphology, only one of the three isolates was found positive for cellulase enzyme. The activity of this isolate at different temperature ranges, different pH and different salt concentrations were determined. In addition, the effects of different carbon sources, organic and inoganic nitrogen sources and different amino acids on enzyme activity were determined. The cellulase produced by this isolate, respectively, at a fixed incubation interval of 48 hours; At 25ºC (48,79 IU/ml) pH 8 (39,78 IU/ml), 5% NaCl (36,07 IU/ml), fructose (37,04 IU/ml), peptone (37,04 IU/ml) and ornithine (36,98 IU/ml) showed high activity in the presence of amino acid. In addition, morphological, physiological and biochemical properties of this isolate were determined. In addition, it was determined that the cellulase positive isolate belonged to Bacillus flexus strain according to 16S rRNA sequencing analysis and its status was determined in bacterial taxonomy.
