Abstract (EN)

In this research, leaf explants from in vivo grown Catharanthus roseus plants were cultured on MS medium supplemented with 5:2; 8:2; 10:2 mg/L NAA:BAP under in vitro conditions. The explants were subcultured once in a four weeks. Callus biomass changing during the 5 subcultures were measured. In addition that, effects of different concentrations of defined plant growth regulators on somaclonal variation were determined. All experiments during the research were realized with three replicates. During the first three subculture series among the subcultures biomass, in the all series generally callus biomass from the beginning of subculture to be end of the first subculture were increased in MS medium supplemented with 5:2; 8:2; 10:2 mg/L NAA:BAP as %168, %139 and %142 respectively. Some such as changes in ploidy level resulted from abnormalities somaclonal variation and metaphase chromosome disorders were defined using the sample squash method from calli. As a result, MS medium supplemented with 5:2 mg/L NAA:BAP were determined as the best combination for biomass production. Furthermore, it was detected that callus biomass increasing changed with different plant growth regulators combination and concentrations and measurements were given as statistically.
