Abstract (EN)

Acute myeloid leukaemia (AML) is one common type of childhood leukaemia and is characterized by the accumulation of abnormal blasts in the bone marrow. In this study, mesenchymal stromal cells derived bone marrow of acute myeloid leukaemia patients and healthy individuals was compared in respect of biological and immunological features. This research was carried out with 7 AML patients, 7 healthy individuals and 1 healthy MNC individual totally 15 individuals. Morphological, population doubling (PD), adipogenic and osteogenic differentiation and cell viability of MSCs were examined. AML patients and healthy individual of MNCs isolated from peripheral blood, T cell activation markers (CD3, CD4, C25, CD69 and HLA-DR) gated positive / positive was analyzed by percent in the flow cytometry apparatus. In the latest experiment, MSCs interact with MNCs that cellular viability was assessed as percent. AML patients with PD value was found to be similar in healthy individuals (z = 1.074; p = 0.394). PHA added and not added to the wells, the results of CD variable values were similar (p> 0.05). Cellular viability value in patients with AML in the wells was determined that there was no difference of statistical significance (x2 = 2,184; p = 0.823). The cell viability values obtained for healthy individuals were similar for all wells (x2= 10,714; p = 0.057). Well 2 is the cell viability values obtained in the healthy individuals groups were statistically significantly higher than the cell viability values obtained in AML patients (z = 2.246, p = 0.026).
